If you finish in the top 5 at a venue and you are unable to compete in the final because you're away on holidays, can you nominate another player eg the 6th placed at your venue; or does NPL Admin do that; or does the spot simply not get filled?

The spot does not get filled. If you are unable to attend it's simply a case of bad luck, sorry. With over 700 qualifiers this season there still won't be a shortage of players.
